Relation between sequence and structure in membrane proteins
MOTIVATION Integral polytopic membrane proteins contain only two types of folds in their transmembrane domains: α-helix bundles and β-barrels. The increasing number of available crystal structures of these proteins permits an initial estimation of how sequence variability affects the structure conservation in their transmembrane domains. We, thus, aim to determine the pairwise sequence identity...
متن کاملThe relation between the divergence of sequence and structure in proteins.
Homologous proteins have regions which retain the same general fold and regions where the folds differ. For pairs of distantly related proteins (residue identity approximately 20%), the regions with the same fold may comprise less than half of each molecule. The regions with the same general fold differ in structure by amounts that increase as the amino acid sequences diverge. The root mean squ...
متن کاملSequence – Structure Relation in Proteins through a “Hybrid Protein Model”
Prediction of protein three-dimensional structures constitutes a major scientific stake. However, it remains difficult even though it is established that the information needed to specify the complex 3D structure of a protein is contained in its amino acid sequence. Our goal is to analyze protein structures at a local level since protein local structural information is encoded to an extent in l...
متن کاملSynthesis of intracellular membrane proteins in vitro. Relation between rough endoplasmic reticulum and mitochondrial outer membrane.
Hepatic rough microsomes were incubated in a messenger-dependent protein-synthesizing system from rabbit reticulocytes. Up to 30% of the total product labelled with [35S]methionine, and subsequently recovered with the microsomes, was located in an intrinsic protein fraction associated with these membranes, i.e. was retained by the membrane following extensive sonication in the presence of 1.5 M...
متن کاملRelation between Protein Structure, Sequence Homology and Composition of Amino Acids Relation between Protein Structure, Sequence Homology and Composition of Amino Acids
A method of quantitative comparison of two classi cations rules applied to protein folding problem is presented. Classi cation of proteins based on sequence homology and based on amino acid composition were compared and analyzed according to this approach. The coe cient of correlation between these classi cation methods and the procedure of estimation of robustness of the coe cient are discussed.
